7-Aminoactinomycin D is a fluorescent DNA stain and RNA polymerase inhibitor

**Background**

The ability to distinguish between viable, apoptotic, and necrotic cells is fundamental in biomedical research, particularly in the study of programmed cell death and oncology. Flow cytometry relies heavily on cell-impermeant dyes to assess membrane integrity, as the loss of membrane selectivity is a hallmark of late apoptosis and necrosis. Furthermore, the inhibition of RNA polymerase is a critical tool for studying gene expression and antimicrobial activity. In the context of cancer research, understanding the transition from early to late apoptosis allows researchers to evaluate the efficacy of various chemotherapeutic agents. Therefore, we will introduce a potent RNA polymerase inhibitor and fluorescent DNA stain – 7-Aminoactinomycin D.

**Definition**

7-Aminoactinomycin D (7-AAD) is a cell-impermeant fluorescent DNA stain and a potent inhibitor of RNA polymerase that selectively binds to GC regions of DNA.

**In Vitro Studies**

According to the 7-Aminoactinomycin D description, this compound exhibits significant biological activity as both a DNA dye and an antibacterial agent. In vitro studies demonstrate its utility in flow cytometry for cell viability assays. Following a recommended 7-Aminoactinomycin D protocol, cells are typically fixed, pelleted, and resuspended before being incubated with 7-AAD (0.5 to 5 µM) for 15 to 60 minutes, with fluorescence measured at Ex/Em = 549/648 nm. At low concentrations (0.5-5 μg/mL), 7-AAD is frequently used to stain and exclude dead cells. At higher concentrations (10-20 μg/mL), it can distinguish between viable cells (7-AAD negative), apoptotic cells (7-AAD dim), and dead cells (7-AAD bright) based on the varying permeability of the cell membrane. Additionally, 7-Aminoactinomycin D biological activity includes the inhibition of RNA polymerase, which disrupts transcription processes. In conclusion, 7-Aminoactinomycin D is a versatile tool for apoptosis detection and the study of RNA polymerase inhibition.
